TNFRSF8
TNF receptor superfamily member 8 (TNFRSF8), also known as CD30, Ki-1 or D1S166E, is a cell membrane protein of the tumor necrosis factor receptor family and functions as the receptor for TNFSF8/CD30L. This protein is expressed by activated, but not by resting, T and B cells. Interacting with TRAF2 and TRAF5, this receptor can mediate the signal transduction that leads to the activation of NF-kappaB and regulation of gene expression. In addition to being a positive regulator of apoptosis, this receptor also plays a role in protecting the body against autoimmunity by limiting the proliferative potential of autoreactive CD8 effector T cells.
This receptor is commonly expressed in various tumors, such as Hodgkin lymphoma (HL), anaplastic large cell lymphoma (ALCL), testicular embryonal carcinoma, therefore, it is widely used as a tumor marker in targeted immune therapy. This receptor is the target of the FDA approved therapeutic brentuximab Vedotin (Adcetris), an antibody drug conjugate (ADC) that is approved for the treatment of patients with HL and systemic ALCL.
