Yeast Hybrid Introduction

Developed from the eukaryotic yeast expression strains, yeast hybrid is a technical approach commonly used for biomolecule interaction tests in vivo. It generally includes: yeast two-hybrid (Y2H) for protein-protein interaction test, yeast one-hybrid (Y1H) for protein-nucleic acid interaction detection, yeast three-hybrid (Y3H) for complex protein interactions, and bacterial two-hybrid (B2H) for protein-protein interaction detection in prokaryotic system. Both the Y1H and Y2H GAL4 systems perform interaction detection by fusing and expressing two candidate genes with two domains of yeast transcription factor GAL4, respectively. Y2H DUAL system is developed based on the split-ubiquitin. Meanwhile, B2H detects protein interactions in E.coli by fusing the proteins of interest with the two complementary fragments of adenylate cyclase, T25 and T18. Additionally, all of these yeast hybrid assays not only can be used for screening interacted candidates in a high-throughput manner but are also often applied for interaction validation of two candidates in different ways.
