Yeast hybrid assays are powerful tools used to study various types of biomolecular interactions in a cellular context. Currently, three types of yeast hybrid assays have been widely used:
Yeast Two-Hybrid (Y2H) Assay: This assay is mainly used to study protein-protein interactions, which is developed by fusing a bait protein to the DNA-binding domain (DBD) of yeast transcriptional factor and a prey protein to an activation domain (AD). When the bait interacts with prey protein, they approach and reconstitute a functional transcription factor that activates reporter genes.
Yeast One-Hybrid (Y1H) Assay: This assay is used to study protein-DNA interactions. It involves a DNA sequence of interest (bait) and a protein of interest (prey) fused to a transcriptional activation domain. When the prey protein binds to the bait DNA sequence, it activates the expression of a reporter gene.
Yeast Three-Hybrid (Y3H) Assay: This assay is used to study RNA-protein interactions. It involves an RNA sequence (bait), a protein of interest (prey) fused to a transcriptional activation domain, and a bridging protein. When the prey protein binds to the RNA bait sequence and the bridging protein interacts with both, it activates the expression of a reporter gene. Moreover, Y3H is also often used to detect the interaction between three proteins, that is, the influence of mediator proteins on the interaction between a pair of proteins.
