Yeast hybrid, especially yeast two hybrid (Y2H), has been a powerful tool for in vivo protein interaction identifications and screening since its discovery in the 1990s. Based on the principle that the yeast Gal4 transcription factor is a protein consisting of two separate but functionally necessary domains, Y2H can screen interacted candidates in a high-throughput manner by constructing fusion expression vectors. Additionally, Y2H is also extensively applied for known protein interaction validation, specific interaction domain determination, and evaluation of drug candidate effects on protein interactions. Owing to the convenient Y2H, several new methods have been developed, such as yeast single hybrid (Y1H), yeast three hybrid (Y3H), and bacterial two hybrids (B2H), largely expanding the detection range to not only detect protein interactions, but also study the interactions between RNA, DNA, and proteins.
