Yao Wang et al. screened DNA aptamers for several commonly labeled proteins and developed an aptamer-based western blotting technique that requires fewer steps and reduces nonspecific binding. Aptamer-based protein recognition analysis was able to selectively label target proteins from complex mixtures and provide a clearer background than conventional antibody-based western blotting. Finally, aptamers are also capable of recognizing fusion proteins carrying these tags, thus greatly expanding the potential applications of such aptamers. This study provides a useful molecular tool for selective protein identification in western blot analysis.
