Applying hybridoma technology in producing monoclonal antibodies is a major breakthrough in biomedicine that has played a huge role in different fields in just a few years. Experimental animals are a necessary condition for the formation of monoclonal antibodies and play an important role in hybridoma monoclonal antibody technology. The selection of suitable hybridoma animals is an important step to produce monoclonal antibodies. Advantages of Animal Immunization

Animal immunization has been used in the production of antibodies for research, diagnostic and therapeutic purposes for more than 120 years, playing an important role in the preparation of antibodies. The use of animal immunization for antibody production has many advantages, including but not limited to:

  1. Animal immunization is a proven and technically mature method.
  2. Antibodies produced by animal immunization undergo in vivo affinity maturation, giving them high affinity and stability in vivo.
  3. High-throughput assays can be used in animal immunization to obtain valuable information about antibody lineage information and paratope patterns.
  4. Antigen acquisition is not necessary to produce antibodies with animal immunization.

Hybridoma Animal Selection

In the process of developing monoclonal antibodies using hybridoma technology, it is often necessary to select appropriate hybridoma animals based on the specific research content and experimental project. To efficiently select appropriate hybridoma animals, several factors need to be considered, including: the species relationship of the antigen to the animal, the individual condition of the animal, and the amount and requirement of the antibody.

Generally, the more distant the source of the antigen is from the phylogenetic relationship between the immunized animal, the more immunogenic its antigen is, and the easier it is to mount an immune response. Conversely, the closer the source of the antigen is to the phylogenetic relationship between the immunized animal, the less immunogenic the antigen is. The age and health of the animal also affect the titer of the antibody produced. Young animals are prone to develop immune tolerance, while elderly animals have a low immune response capacity and are often unable to produce high-titer antibodies. Therefore, it is better to use healthy animals of appropriate age, strength, and weight antibody preparation. Special attention should also be paid to nutrition and hygiene management during immunization. Finally, the type of hybridoma animal should be determined based on the required use of the antibody and the amount of antibody needed.
