Assay Description
Electric cell-substrate impedance sensing (ECIS) is an impedance-based technique used for real-time monitoring of cellular events such as cell migration, invasion, proliferation and activities of cytoskeleton. Cells are grown on special types of electrodes (gold electrodes) and electrodes are attached in the bottom of culture wells. Impedance vs. time is measured upon supplying an AC current during the experiment and due to the dielectric nature of the cell membrane impedance, increases as cells form a layer on electrodes. The three-dimensional (3D) shape of cells is the main factor for impedance as the changes in cell shape/structure cause increase or decrease in impedance. Therefore, real-time monitoring of cell proliferation based on impedance is possible with this technique. As the shape and cell adhesion to a surface alter upon treat ment with cytotoxic compounds, this technique can also be applied to monitor effects of a test drug on cell shape alterations. The Cell Index is a unit that describes the relationship between electronic impedance and viable cell number. Chemotactic and chemokinetic properties of components can also be monitored with this technique.
