Assay Description
Serial analysis of gene expression (SAGE) is another molecular biological technique used to analyze gene expression. In this technique, mRNA extracted from the sample is reverse transcribed into cDNA and the resulting cDNA is then digested by restriction enzymes to produce tags. These tags are amplified by PCR and subjected to sequencing. Several modifications to SAGE such as Long SAGE, RL-SAGE and Super SAGE have been introduced to improve the SAGE technique.

Following transcription, primary transcript RNA is converted into mature RNA by post-transcriptional modifications. Post-transcriptional modification includes three major steps namely the addition of a 5'cap, addition of a 3'polyadenylation tail and RNA splicing. Stability of RNA has been considered as very important for gene expression and proteins at the post-transcriptional level.
