Magic™ Cell-free Protein Expression in Mammalian Cell Lysate Kit is a cell-free protein synthesis system using human cell extracts. This system contains all the required components for protein synthesis (ribosomes, translation initiation /elongation /enhancement factors, tRNA, polymerase, amino acids, ATP etc.) and allows protein synthesis to occur at high efficiency and high yield. Modifications such as glycosylation and phosphorylation could be easily obtained. The procedure can be performed in a single reaction tube from RNA transcription to protein synthesis.
The transcribed target gene RNA contains an IRES sequence designed to promote protein translation initiation. Although the translation initiation factor in the cell extract becomes inactivated as protein synthesis progresses, it can be reactivated by the translation enhancement factor included in the Accessory Proteins, therefore maintaining the translation level.
