Nipah Virus

Nipah Virus (NiV), belonging to the member of genus Henipavirus in the family Paramyxoviridae, is an enveloped virus, pleomorphic in shape (40-1900 nm) with a single-stranded negative-sense RNA genome and causes serious disease in both animal and human populations characterized by a respiratory and/or acute encephalitis syndrome with high mortality.

Live Attenuated Nipah Virus Vaccine

The best standard for assessing the efficacy of a vaccine against a virus that causes hemorrhagic fever is a non-human primate model that recapitulates human disease. African green monkeys (AGM) have been used as models to establish the Nipah virus, and the disease in this model is very similar to the disease of human disease. Nipah virus is highly toxic in this model and causes severe respiratory diseases. Live-attenuated vesicular stomatitis virus vaccine that encodes the Nipah virus glycoprotein as an antigen can protect AGM. Vaccination produces a rapid and strong viral-specific immune response that inhibits virus shedding and replication.

Viral Vector based Nipah Virus Vaccine

Based on existing knowledge and studies on the immunity of other paramyxoviruses, NiV envelope proteins F (fusion) and G (glycoprotein) were found to be used for vaccine development. The recombinant NiV F and G proteins expressed by vaccinia virus are immunogenic and can induce a protective immune response in hamsters. Canarypox virus-based vaccine vectors carrying NiV F or G proteins induce neutralizing antibodies in pigs and prevent viral shedding during NiV challenge.

Subunit Nipah Virus Vaccine

Both NiV and Hendra virus (HeV) are paramyxoviruses but have different biological, molecular, and serological properties than all other paramyxoviruses, and therefore, they are grouped together as closely related viruses in the new Henipavirus genus. Subunit vaccine composed of the G Glycoprotein of HeV virus can protect AGMs from Nipah virus challenge.

Peptide or Epitope Based Nipah Virus Vaccine

The peptide or epitope-based vaccines are easier to produce, more specific, more cost effective, less time consuming, and safer than conventional vaccines. T cells play a key role in inducing cellular immune responses against foreign antigens, and they recognize antigenic fragments only when they are associated with major histocompatibility complex (MHC) molecules that are exposed on the surface of all vertebrate cells. Immunoinformatics methods use computational algorithms to predict potential candidate vaccine or T cell epitopes. An advantage of a peptide or epitope-based vaccine is to deliver high doses of potential immunogens at low cost. Viral proteins that can be used as vaccine candidates must be surface exposed and antigenic.
