Thermal Stable mAbs are Needed in Clinical Therapy
Antibodies and their derivative fragments have long been used as tools in a variety of applications, in fundamental research work, biotechnology, diagnosis, and even human therapy. Not surprisingly, the composition of antibodies in clinical trials is increasing. For most applications, besides the need of efficient biodistribution or reduced immunogenicity, thermals stability and solubility are critical factors. The benefits of producing thermal stable mAbs are that they typically express at high levels in Escherichia coli and remain soluble in solution at higher concentrations for longer periods of time.
Thermal Stable mAbs Discovery in Creative Biolabs
In terms of the extensive experience in antibody research, Creative Biolabs has developed a variety of strategies for the identification of thermal stable mAbs. These strategies take advantage of the fact that bacteriophage M13 particles are heat stable. An antibody library is expressed on the surface of bacteriophage M13 when under high temperature, thermal unstable clones show reduced or no binding activity and will be lost during subsequent affinity selection. However, thermal unstable clones that still retain binding to their target are selected and enriched with each subsequent round of affinity selection. In order to confirm that elevated temperatures are effective in eliminating thermal unstable phage clones, the phage library is heated at various temperature. Effect of high temperature on eliminating thermal unstable clones can be tested by phage ELISA. After the identification of thermal stable phage clones, the melting temperature (Tm) will be determined.
