Background

DNA methyltransferase 1 (DNMT1) & Sonic hedgehog protein (SHH) are both attractive targets in cancer research. DNMT1 is an enzyme responsible for catalyzing DNA methylation. It is related to tumorigenesis of various types of cancers, including leukemias, lymphomas, breast cancer, liver cancer, etc. Meanwhile, SHH is a member of hh homologues encoding SHH protein, whose abnormal activated signaling in adult tissues is implicated in a wide range of cancers, such as skin, breast, brain, gallbladder, and liver cancer. It is reported that silencing of DNMT1 suppresses the HH pathway, leading to the inhibition of epithelial-mesenchymal transition (EMT) in liver fibrosis. With the ability to simultaneously inhibit DNA methylation and HH-PTCH receptor signaling, anti-DNMT1×anti-SHH bispecific antibody (BsAb) conjugated to vinorelbine-tartrate is demonstrated to result in the re-expression of apoptotic tumor suppressor genes, eliminating the therapy resistance in hormone refractory breast cancer.
