Aiming to discover antibodies with better performance in a short period, Creative Biolabs currently has pushed out a high-throughput B cell sorting platform, which is capable of screening a large number of specific B-cells from humans or immunized animals, and also identifying rare antibodies with the biological activity of interest and favorable manufacturing properties.

To obtain antibodies with naturally paired heavy chain and light chain, the single B cell antibody technology is preferred, which is capable of high-throughput screening and generating natural monoclonal antibodies (mAbs) of different species in a short period. Firstly, humans or animals should be properly immunized to activate naive B cells to differentiate into antibody-secreting B (ASB) cells. Next and importantly is antigen-specific single B cell isolating and identifying, which is the key to the whole process. Single B cell isolation from peripheral blood or lymphoid tissues usually is divided into two scenarios according to different research purposes or applications, one is random B cell isolation and another is antigen-specific B cell identification.

Single B cell random isolation is performed by directly picking the B cells from the sample, which is suitable for blood samples with a high concentration of antigen-specific antibodies. This random isolation usually is applied for single B cell isolation from vaccinated individuals or patients with a single infection. Unlike random isolation, antigen-specific single B cell isolation is selecting the target-specific ASB cells from pre-immunized samples, which is widely used and is especially preferred when it comes to low antibody concentration in samples, such as anti-tumor antibodies, autoimmune antibodies, and rare antibodies.
