Gal-3 is a 29- to 35-kDa protein that in humans is encoded by the LGALS3 gene located on chromosome 14. And it belongs to the member of the lectin family of which 15 galectins have been identified in mammals. As a family of β-galactoside-binding lectins with ≥1 evolutionary conserved carbohydrate-recognition domain (CDR), galectins are divided into three types based on its domain organization, which are prototype galectins with one single CDR, tandem-repeat galectins with two CDRs and chimera-type galectins with a single CDR connected to a long, flexible N-terminal domain. And galectin-3 is the only chimera-type galectin which binds proteins in a carbohydrate-dependent and -independent manner.
The Role of Gal-3 in Tissues
Gal-3 is widely expressed in human tissues which contain all types of immune cell (monocytes, mast cells, natural killer cells, dendritic cells, macrophages, eosinophils, and activated T and B cells), endothelial cells, epithelial cells, and sensory neurons. The expression of Gal-3 is more abundant in embryogenesis and development than adult life, which gets developmentally regulated. This protein is predominantly either situated in the cytoplasm and transported into the nucleus or secreted toward the cell surface and converged to the biological fluids. The various functions of Gal-3 depend on where it is located. The fact that Gal-3 in the cytoplasm interact with certain survival-associated proteins like B-cell lymphoma-2 (Bcl-2) and activated guanosine-5'-triphosphate (GTP)-bound K-Ras makes it play an essential role in cell survival. Gal-3 in the nucleus promotes pre-mRNA splicing and regulates gene transcription whereas extracellular Gal-3 modulates cell-cell interactions including between epithelial cells and the extracellular matrix.
Gal-3 as a Novel Diagnostic Biomarker
Gal-3 has the potential to be used in the diagnosis and prognosis of various types of disease and serve as a therapeutic target for treating disease owing to that it is a stable biomarker and is not affected by age body mass index or sex. Several studies have concentrated their much energy on the role of Gal-3 in the models of metabolic disorders such as experimentally induced diabetes, AGE injection, the normal aging process and feeding a high-fat diet (HFD). It has been demonstrated that Gal-3 plays a pivotal role in the regulation of the molecular processes underlying injury in all organs and tissues that are affected by complications of these conditions. Mechanism of action of Gal-3 in many disorders including nephropathy, retinopathy, atherosclerosis and liver disease has been revealed.
