Mycosis generally refers to infectious diseases caused by pathogenic fungal infections, which can be generally divided into two types, opportunistic fungal infections and primary fungal infections. The emergence of drug-resistant fungi has brought more severe challenges and broad development space for antifungal drugs development. Determining the mechanism of action of antifungal compounds is critical for the discovery of antibiotics, but its complexity largely limits the development of basic research and antifungal discovery programs. Fungal cytology (FCP) is a rapid and efficient method to identify cellular pathways affected by antimicrobial molecules.
Although cytological assays were originally developed in eukaryotic systems, studies have shown that this assay method can also be used to gain insights into the mechanism of action of antifungal drugs in yeast. This platform enables us to rapidly characterize the MOA of candidate molecules in the early stages of antifungal drug development to expedite the drug development process and minimize the loss of project failures.
We have verified the effectiveness of this method in various strains including C. Albicans, A.fumigatus, as well as C.neoformans. In this assay, the antifungals would incubate with the fungal strains of interest at 35°C with shaking. At 4 hours and 24 hours, treated cultures will be stained and viewed under a high-resolution fluorescence microscope, followed by quantification of various cytological parameters to create a cytological map. This method can rapidly and accurately distinguish whether antifungal agents inhibit specific biosynthetic pathways and sub-pathways. What's more, it can also reveal if the antifungal agent is affecting multiple pathways.
