PAD is characterized by the blocking of the arteries in the lower extremities, failing to transport enough blood or oxygen from the heart to other parts of the body. As a common arterial disease in the elderly, PAD often causes discomfort or pain when walking. Smoking and diabetes can greatly increase the risk of PAD. Clinical evidence shows that the arterial lumen becomes narrow or even completely occluded in PAD patients with reduced blood flow. PAD may not show obvious symptoms but requires timely treatment since it can easily induce heart disease and stroke.
Animal disease models are important tools for understanding disease pathogenesis and progression and evaluating the efficacy of potential therapies. The commonly used PAD models are the bilateral laurate-induced PAD model and femoral artery ligation model in rats. The classical laurate-induced PAD model has been used to evaluate anti-thrombotic agents. In addition, there are currently emerging computer simulations, mathematical models, or in vitro models combined with in vivo models to reduce the number of animals required for experimental research.
It is known that NHPs are the experimental animals closest to humans in terms of anatomy, genetics, physiology, and biochemistry. Although NHPs are larger and more expensive than rodents, their advantages as preclinical disease models make them an optimal choice for more and more researchers.
