Background
Similar to PAR1, PAR2 is activated by proteolytic cleavage of its N-terminus by serine proteases. This cleavage exposes a new N-terminus, which acts as a tethered ligand that binds to and activates the receptor. PAR2 is widely expressed in various tissues including epithelial cells, neurons, immune cells, and vascular endothelial cells. It plays a significant role in mediating inflammatory responses, pain sensation, gastrointestinal function, and skin biology.
