The method developed by Puchtler remains the gold standard for amyloid in tissue sections. Amyloid clusters are birefringent and appear green in polarized light.
Description
The Congo Red Puchtler kit is used to stain amyloid (amorphous clusters) in tissue sections. Despite their differences in protein composition, amyloid deposits are associated with fibrillar beta sheet formation. Congo red dye binds to fibrils and forms a regular pattern of dye molecules in the process. The structural regularity of the dye under polarized light shows the birefringence of light by showing green color. According to Puchtler, the high pH level and ionic strength of the solution allow for higher dye specificity for amyloid.
