HEK293-Tg(Human BCMA)-NF-κB reporter cell line is a cell line derived from HEK293 cells that has been genetically modified to stably express the human BCMA (B-cell maturation antigen) gene along with an NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) reporter gene. This cell line is commonly used in research to study the signaling pathways activated by BCMA and how it influences NF-κB activation. The NF-κB reporter gene allows for easy monitoring of NF-κB activity in response to BCMA stimulation.

The BCMA (B cell maturation antigen) gene is located on chromosome 16q23 and encodes a cell surface protein that is a member of the tumor necrosis factor receptor superfamily. BCMA is primarily expressed on the surface of plasma cells and activated memory B cells, and it plays a crucial role in the survival and function of these cells. BCMA interacts with its ligands BAFF (B cell activating factor) and APRIL (a proliferation-inducing ligand) to promote the survival and differentiation of plasma cells, as well as regulate the immune response. Dysregulation of BCMA signaling has been associated with various autoimmune diseases and cancers, making it a potential therapeutic target for these conditions. Mutations in the BCMA gene have been linked to multiple myeloma, a type of cancer that affects plasma cells, as well as other B cell-related disorders. Targeting BCMA with monoclonal antibodies and chimeric antigen receptor (CAR) T-cell therapies has shown promising results in the treatment of multiple myeloma and other B cell malignancies.
