Description
The Jurkat-Tg(Human TIGIT)-NFAT Luc reporter cell line is a cell line derived from Jurkat T cells that have been genetically modified to express human TIGIT (T cell immunoreceptor with immunoglobulin and ITIM domains) protein. This cell line also contains a luciferase reporter gene under the control of NFAT (nuclear factor of activated T cells) response elements, allowing for monitoring of NFAT activity. The expression of human TIGIT in this cell line allows for the study of TIGIT-mediated signaling pathways and immune responses. The NFAT luciferase reporter provides a readout of NFAT activation, which is a key transcription factor involved in T cell activation and differentiation.

Background
The TIGIT gene is a gene that encodes a protein known as T cell immunoreceptor with immunoglobulin and ITIM domains. TIGIT is a receptor expressed on the surface of T cells, particularly regulatory T cells, and plays a role in regulating immune responses. TIGIT interacts with its ligands CD155 and CD112, which are expressed on antigen-presenting cells and tumor cells, to modulate T cell activation and function. TIGIT has been implicated in immune tolerance, inhibiting T cell responses, and promoting immune evasion by tumor cells. Targeting TIGIT with therapeutics is being explored as a potential strategy for cancer therapy.
