Three-reagent kit for collagen staining and visualization in histology. Contains Sirius Red dye that enhances collagen birefringence so it can be easily identified by viewing sections under polarized light.

The Picro-Sirius Red kit is used for visualization and collagen staining of formalin-fixed and paraffin-embedded tissues. It contains Sirius Red dye used in histology to visualize collagen fibers. Strong linear anionic dyes stain collagen at low pH by reacting with bases on collagen molecules. Sirius Red binds in parallel with collagen molecules; it also enhances the birefringence of collagen so that it can be easily identified by viewing sections under polarized light. The kit contains Eriochrome cyanine R dye, which counterstains cell nuclei to a gray-brown color. Eriochrome is acid-fast; prevents discoloration of nuclei after prolonged incubation in Picro-Sirius Red solution.
