- Overview
- Reticulon 4, also known as neurite outgrowth inhibitor or Nogo, is a protein encoded by the RTN4 gene in humans and has been identified as a neurite outgrowth inhibitor unique to the central nervous system . During nerve development, Nogo is mainly expressed by neurons and provides inhibitory signals for the migration and germination of CNS endothelial (tip) cells, thereby limiting the density of blood vessels. There are three isoforms: Nogo A, B and C. Nogo-A has two known inhibitory domains, including the N-terminal amino Nogo and Nogo-66, the latter constituting the molecular extracellular loop. Both Amino-Nogo and Nogo-66 are involved in the inhibition reaction, of which Amino-Nogo is a strong inhibitor of neurite outgrowth, and Nogo-66 is involved in the destruction of the growth cone.
Studies have shown that blocking Nogo-A during neuronal damage (from diseases such as multiple sclerosis) can help protect or restore damaged neurons. [9] The study of the mechanism of this protein provides great potential for the treatment of autoimmune-mediated demyelinating diseases and spinal cord injury regeneration. It has also been found that it is a key participant in this process, and physical exercise can enhance the learning and memory processes of the brain. also shows that Nogo-A has a negative regulatory effect on blood vessel growth and repair after ischemic stroke. In the experimental stroke mouse model, Nogo-A gene deletion and antibody-mediated blockage resulted in enhanced revascularization and functional recovery. In addition, anti-Nogo-A antibody treatment reduces vascular leakage, the main complication after stroke.
