In recent decades, research focuses on ATM distribution, cytokines involved in ATM regulation, ATM inhibitors and so forth. Creative Biolabs provides one-stop services with a variety of test platforms for ATM analysis. Currently, we have launched a series of biomarker analysis services to promote disease research. Here, we introduce the ATM biomarker analysis.
ATM serine/threonine kinase (ATM) activation occurs in two different ways. The classic ATM activation pathway relies on double-strand DNA break (DSB) and MRN complexes. ATM usually exists in the nucleus as a homodimer (non-activated state), which becomes a monomer after activation to interact with the end of DNA and its substrate, and then participates in cell cycle checkpoint control, apoptotic reaction and DNA repair, etc. The other way is to form disulfide bond between two ATM monomers under oxidative stress, thus activating ATM.
ATM activity has been associated with a variety of diseases and can be monitored in real time using traditional protein analysis methods, fluorescent protein and in vitro bioluminescence techniques.

Loss of ATM protein activity in the human body leads to a polytropic neurodegenerative disease called ataxia telangiectasia (AT), which leads to immune system deficiencies, cancer susceptibility, etc.
Recent studies have shown that ATM can repair DNA DSBs and that targeted therapy enables tumor cells to become constitutionally dependent on ATM kinase. ATM inhibitors can clear resistant residual tumor cells, resulting in a more permeable, longer lasting effect. Some ATM inhibitors have emerged as potential candidates for cancer treatment, the following are several ATM inhibitors.
CGK733
ATM/ATR inhibitors, used in cancer research.
AZ32
An orally available ATM inhibitor that penetrates the blood-brain barrier.
CP-466722
Reversible ATM inhibitors have no effect on PI3K and PIKKs family proteins.
Ro 90-7501
Inhibit ATM phosphorylation and DNA repair.
In the past few years, significant advances have been made in ATM molecular-specific protein assays, fluorescence or radiolabeling.
