Cluster of Differentiation 74 (CD74)
HLA class II histocompatibility antigen gamma chain, also known as HLA-DR antigen-associated invariant chain or CD74 (Cluster of Differentiation 74), is a protein that in humans is encoded by the CD74 gene. The invariant chain is a polypeptide involved in the formation and transport of MHC class II protein. The cell surface form of the invariant chain is known as CD74. The protein encoded by CD74 gene associates with class II major histocompatibility complex (MHC) and is an important chaperone that regulates antigen presentation for immune response. It also serves as cell surface receptor for the cytokine macrophage migration inhibitory factor.
CD74 is an integral transmembrane molecule playing a role in intracellular sorting of MHC class II molecules. This molecule is expressed by most of the B-cells, particularly follicular center cells, mantle cells, and activated B-lymphocytes. A subpopulation of T-cells, macrophages, activated endothelial cells, and neoplastic plasma cells may also express CD74. CD74 is a cell-surface receptor for the cytokine macrophage migration inhibitory factor. Macrophage migration inhibitory factor binding to CD74 induces its intramembrane cleavage and the release of its cytosolic intracellular domain (CD74–ICD), which regulates cell survival.

CD74 Marker of Lymph Cancer
It is found that CD74 are expressed on a number of cancer cell types. CD74 has been shown to regulate peripheral B cell survival and is associated with tumor progression and metastasis. The CD74 expression has been suggested to serve as a prognostic factor in many cancers, with higher relative expression of CD74 behaving as a marker of tumor progression. Previous studies showed that stimulation of CD74 expressed on chronic lymphocytic cells initiates a signaling cascade leading to chronic lymphocytic survival.
