Survivin

Survivin, also called baculoviral inhibitor of apoptosis repeat-containing 5 (BIRC5), is a member of the inhibitor of apoptosis family. The survivin protein is encoded by the BIRC5 gene in human. The functions include inhibiting caspase activation, thereby leading to negative regulation of programmed cell death or apoptosis. This has been shown by disrupting the survivin-induced pathways, leading to increased apoptosis and reduced tumor growth. The expression of survivin is also highly regulated by the cell cycle and is expressed only in the G2-M phase. Survivin is known to be localized to mitotic spindles by interacting with tubulin during mitosis and may play a contributing role in the regulation of mitosis. The molecular mechanism of survivin regulation is unclear, but the regulation of survivin appears to be related to the p53 protein.

Survivin as Marker of Prostate Cancer

Survivin is expressed in embryonic and foetal tissues but is undetectable in normal adult tissues. However, survivin overexpression has been reported in almost all human malignancies, including prostate, breast cancer, ovarian cancer, lung cancer, gastric cancer, liver cancer, bladder cancer, and hematological cancers. Expression levels of survivin are associated with more aggressive disease and poor clinical outcome. It is minimally expressed in normal tissues, so it has become a major target for tumor in vitro diagnosis. Besides, overexpression of survivin in cancer can overcome cell cycle checkpoints to promote abnormal progression of transformed cells through mitosis. What's more, survivin has not been found to be expressed in normal secretory epithelium of the prostate but has been found to be strongly expressed in prostate cancer cells. The differential expression of survivin in prostate cancer cells compared to normal tissues make it a highly promising target for diagnostics of prostate cancer.
