Collagen Antibody Induced Arthritis (CAIA) Model

Rheumatoid arthritis (RA) is a typical, chronic autoimmune disease that primarily affects the joints, ultimately leading to synovitis that often develops into cartilage and bone erosion, apart from which, extra-articular features are presented, including subcutaneous nodules, vasculitis, and pulmonary fibrosis, particularly in severer cases. Collagen antibody-induced arthritis (CAIA), as a widely used model of arthritis induced by systemic administration of mixtures of CII-specific monoclonal antibodies that target various epitopes of type II collagen, is the direct derivative of CIA. Conventionally, a cocktail of arthritogenic monoclonal antibodies directed against type II collagen (CII) combined with a boost of LPS is used to induce arthritis in CAIA model. Practically, this model is useful for studying the pathological role of individual gene products, pro-inflammatory and anti-inflammatory cytokines, screening and evaluating anti-inflammatory agents. Advantages of Collagen Antibody Induced Arthritis (CAIA)

As is seen in CIA, CAIA model displays similar clinical and histologic characteristics and simultaneously involves important features of human RA, such as inflammatory synovitis, formation of pannus (an aggressive fibrovascular tissue that invades the joint), cartilage destruction, and bone resorption, although acute components (e.g. infiltration with neutrophils) prevail when compared to CIA.

One particular advantage of CAIA over CIA is that the model is amenable for use in strains or genotypes not genetically susceptible for CIA. Moreover, CAIA, in contrast to CIA, is a rapid model (onset within 48h plus peak disease within 8 days) with high uptake rate, synchronicity and lack of influence of Freund's adjuvant, which was used for CIA model and affects the immune system. CAIA requires formation of immune complexes on the cartilage and complement activation while its induction of arthritis is B cell and T cell independent.
