CAR Design Construction and Plasmid GLP Production
Creative Biolabs develops and designs chimeric antigen receptors (CAR), to detect and fight cancer cells. This protein structure can
stimulate cancer-fighting T cells and thus strengthen the patient's immune system. Relies heavily on historical

CAR Designs Overview
Creative Biolabs provides four types of CAR for experimental research, making your research on cellular gene therapy
more effective and convenient.
√ 1st-generation CAR
It contains CD3ζ-chain and the primary transmitter of signals from endogenous TCRs, which show success in
pre-clinical trials and entered Phase I clinical trials in ovarian cancer, neuroblastoma and various types of
leukemia and lymphoma.
√ 2nd -generation CAR
The co-stimulatory domain is derived from either CD28 or 4-1BB, which are co-stimulatory receptors expressed on the
surface of T cells. And the CD28-based CARs show rapid proliferative response and boost effector functions.
Meanwhile, the 4-1BB-based CARs manifest a more progressive T cell accumulation.
√ 3rd-generation CAR
Incorporate two co-stimulatory domains, such as CD3 ζ-CD28-41BB, CD3 ζ-CD28-OX40, both are derived from different
co-stimulatory proteins, which can enhance further activation signals, proliferation, production of cytokines and
effective function.
√ 4th-generation CAR
The CAR is designed to shape the tumor environment by the inducible release of transgenic immune modifiers, such as
IL-12, which augments T-cell activation, attracts and activates innate immune cells to eliminate antigen-negative
cancer cells in the targeted lesion.
