Background
GPRC5A was identified and initially characterized through genomic sequencing and bioinformatics approaches aimed at cataloging GPCRs. It belongs to the Class C (metabotropic glutamate receptor-like) family of GPCRs, which are characterized by a large N-terminal extracellular domain and a long cysteine-rich domain involved in ligand binding. GPRC5A, like other Class C GPCRs, consists of an extracellular N-terminal domain, seven transmembrane helices connected by intra- and extracellular loops, and an intracellular C-terminal domain. The N-terminal domain of GPRC5A is notable for its cysteine-rich region, which is involved in ligand binding and receptor activation. GPRC5A is widely expressed in various tissues throughout the body, with higher expression levels observed in the brain, lung, kidney, and liver. Its expression pattern suggests potential roles in diverse physiological processes, including sensory perception, cellular signaling, and metabolic regulation.
