Cytokines Regulation in the Gut-Brain Axis

The gut microbiome is an integral part of the gut-brain axis. The central nervous system (CNS) and the gut microbiota interact in a bidirectional manner. These close interactions involve endocrine, neurological, metabolic, and immunological communication to monitor and influence gut and CNS function. Gut microbiota and their metabolites play an important role in regulating inflammatory responses in the CNS. A variety of biologically active products derived from gut microbiota can directly or indirectly influence the brain, causing the release of inflammatory cytokines by microglial cells within the CNS, or indirectly by inducing the release of inflammatory cytokines from the gastrointestinal (GI) tract. Growing experimental and clinical evidence suggests that a chronic inflammatory response induced by the gut microbiome can contribute to the development of neuro diseases such as autism spectrum disorders (ASDs).

Strategies for Cytokine Analysis

ELISA is the time-tested and best-validated method for measuring individual cytokines. The inability of traditional ELISA to multiplex presents additional limitations in sampling volume, cost, and labor. A multiplex cytokine analysis can allow researchers to investigate analytes simultaneously, saving time, samples, and resources. We offer a broad range of multiplex immunoassays, including membrane antibody arrays, glass-slide-based quantitative antibody arrays, and suspension arrays. The planar arrays are of two types, either the slide or microtitre-based format. In planar microassays, the capture ligands are immobilized on a rigid two-dimensional support, probed with a sample and the fluorescent or chemiluminescent signals detected. The suspension array utilizes fluorescently labeled magnetic microparticles or beads conjugated to specific-capture antibodies to detect up to 100 different analytes simultaneously.
