TCR/CAR Expression Analysis
Evaluating the expression of TCR/CAR is an essential step in the production of TCR-T & CAR-T cells. This is usually done by flow cytometry using protein L, anti-Fab antibodies or target antigens as detection antibodies. Creative Biolabs has extensive experience in evaluating TCR/CAR expression analysis. With a strong team of Ph. D. scientists and state-of-the-art analytical instruments, we are able to perform TCR/CAR expression analysis in the most time- and cost-effective manner.
Creative Biolabs offers a range of validation assays that span the entire process of TCR/CAR engineered T cell therapy, such as TCR/CAR gene expression at various stages after TCR gene transfection, cell preparation and clinical treatment. Therefore, classical measurements include Western blot (WB), enzyme-linked immunosorbent assay (ELISA), flow cytometry (FCM), immunohistochemistry (IHC), etc., which can be integrated according to customers’ purpose. We can provide reliable data and comprehensive analysis reports, including detailed procedures and appropriate charts and calculations.
TCR/CAR Gene Expression Analysis
We isolate total RNA from T cells following the instructions of the RNeasy kit. The expression of the target gene is screened using a human immunology kit and analysis software. Genomic Suite is used to analyze gene expression changes. For real-time polymerase chain reaction (PCR) analysis of gene expression, we use a pathway-focused T cell energy and tolerance RT2 Profiler PCR array. The genes analyzed by PCR arrays are further used to measure target gene expression levels with our designed TaqMan probe/primer set.
TCR/CAR Protein Expression Analysis
To assess TCR/CAR protein expression levels, Creative Biolabs provides a series of validation analyses to determine the expression levels of TCR/CAR genes throughout the course of engineered T cell therapy. Classical measurements include Western blot (WB), enzyme-linked immunosorbent assay (ELISA), flow cytometry (FCM), and immunohistochemistry (IHC)
In general, the most common and effective methods we use is flow cytometry (FCM). At Creative Biolabs, we perform analytical flow cytometry analysis with FACSDiva software and multicolor sorting and analysis using FlowJo software. Negative selection of T cells is performed using a T cell isolation kit. Antibodies can be selected from our products or supplied by the customer.
