Introduction to ANGPT2

ANGPT2 (Angiopoietin-2), a protein encoded by the ANGPT2 gene in humans, is a member of the angiopoietin family, which is synthesized, stored, and secreted by endothelial cells (ECs). ANGPT2, which has been reported to be abundantly expressed in a variety of tumor cells and to play a significant role in tumor angiogenesis and inflammation, has been identified as an appealing target for antiangiogenic therapy. ANGPT2 was formerly thought to be a naturally occurring antagonist of ANGPT1 because it can compete with ANGPT1 for binding to TIE2, lowering TIE2 phosphorylation in vascular ECs, which might impair vascular stability and promote angiogenesis, and may impart resistance to antiangiogenic therapy. However, ANGPT2 can potentially operate as a mild agonist to stimulate vascular remodeling in adult tissues. Furthermore, ANGPT2 is expressed in endothelial cells of dynamically remodeling arteries, such as tumors, and contributes to endothelial instability in inflammation and cancer.

ANGPT2 for Cancer Diagnosis & Prognosis

ANGPT2 plays an important role in tumor angiogenesis in Hepatocellular carcinoma (HCC), ANGPT2 is highly expressed in HCC, and ANGPT2 levels are strongly linked to HCC development and prognosis. ANGPT2 is also discovered to be a unique predictive biomarker for overall survival (OS) in advanced gastric cancer that is substantially related to liver metastasis (LM). Furthermore, studies have identified links between Angpt2 polymorphisms and CRC development and progression in Chinese Han people. There is evidence that serum ANGPT2 level has potential usefulness for early identification of breast cancer and has prognostic significance in breast cancer patients. Higher level of ANGPT2 is associated with higher tumor stage in lung cancer patients with solitary pulmonary nodule (SPN), indicating that serum ANGPT2 measurement can be a valuable diagnostic tool for SPN patients. It is claimed that combining VEGF inhibition and ANGPT2 suppression may overcome drug resistance to glioma treatment. ANGPT2 has also been identified as a biomarker for both diagnosis and prognosis in individuals with metastatic melanoma receiving immune checkpoint treatment.
