Introduction and Function of Chemokine CCL
CCL chemokines, with an N-terminal CC domain, are a family of more than 20 chemotactic cytokines. They are an important component for communication between cells and tissues, which is crucial for the functioning of the homeostasis of the immune system and tumor development.
The physiological function of CCL chemokines, produced by tumor cells and tumor-associated cells, includes the recruitment of various cells, such as regulatory T cells (Treg), tumor-associated macrophages (TAM), myeloid-derived suppressor cells (MDSC), tumor-infiltrating lymphocytes (TIL) and other immune cells. As shown in Fig.1, CCL chemokines promote the proliferation, migration, and invasion of cancer cells. At the same time, they also play a key role in the angiogenesis and lymphangiogenesis processes, which are essential for organ-specific metastasis of tumor cells. Various CCL chemokines have been reported to be associated with drug resistance in cancer immunotherapies, including the Engineered Cancer Cells with the self-destruction strategy.

Measure Chemokine CCL Production by FC
The FC assay allows the measurement of chemokine CCL production by defined cell populations. In the flow cytometry experiment, after cell stimulation, regents are added to prevent chemokine secretion from the cells. Then cells will be fixed, permeabilized, intracellular stained with antibodies against chemokine CCL, and finally detected by Flow Cytometer. The lab of Creative Biolabs is equipped with a Flow cytometer which has multiple lasers and fluorescent channels. Our experienced experts will do cell sorting or cell analysis needed with limited cell subpopulations for the measurement of chemokine CCL production.

Chemokine CCL Production Measurement by ELISA
ELISA can be used to measure the production of CCL chemokines that are secreted into the supernatant by stimulated cells. ELISA is more sensitive than FC, while could not identify the cells producing chemokine. As for the ELISA procedure, 96-well plates will be incubated overnight with the capture Antibody. The next day, block the wells' surface sufficiently to prevent non-specific bindings, then add the sample or chemokine standards, and finally, the labeled secondary antibody will detect the existence of chemokine. Between these steps, we need to wash plates to remove the background. In the end, we read the absorbance on a spectrofluorometer. Chemokine CCL Production Measurement by QPCR
QPCR assay has been developed for immunological investigations of the gene expression of human chemokines in infected or stimulated cells. QPCR could help us understand the origin and functions of chemokine-producing and assess the immune system and identify ideal disease biomarkers. As for the QPCR procedure, samples will be collected to extract RNA for the following reverse transcription. QPCR will run with specific primers we designed for interested chemokine genes.
