Breast Cancer Cell HCC1937
Predisposition to breast cancer may arise from germline mutations in either of two tumor suppressor genes, BRCA1 and BRCA2. Mouse embryos that lack wild-type BRCA1 or 2 functions undergo early embryonic arrest which is most likely associated with the activation of cell cycle checkpoints. Besides, primary cells from such embryos reveal spontaneous chromosomal anomalies in the absence of extrinsic DNA damage. Furthermore, murine BRCA1-induced tumorigenesis is promoted by the preexisting inactivation of p53.
HCC1937, the BRCA1-mutated breast cancer cell line, synthesizes a truncated BRCA1 protein that is the product of a disease-producing mutant allele (5382insC). Grade 3BRCA1 analysis revealed that the HCC1937 cell line is homozygous for the BRCA1 5382C mutation. HCC1937 cell line is positive for the epithelial cell-specific marker Epithelial Glycoprotein 2 (EGP2) and for cytokeratin 19, and negative for expression of estrogen receptor (ER) and progesterone receptor (PR).
