T Cell-based Multimer/Tetramer Assay

Antigen-specific T cells existing in peripheral blood can respond to antigen infection quickly, and the mechanism is based on the T cell receptors (TCR) on the cell surface. Inspired by the unique binding interaction between TCR and peptide-MHC, Creative Biolabs offers synthesized peptide-MHC multimer/tetramer and its application in cancer epitope analysis.

Peptide-MHC Tetramer/Multimer

The exquisite specificity of T cell response to peptide/MHC is useful in tracing antigen-specific T cells and analyzing antigenic peptides. But the binding affinity of a single peptide/MHC to its cognate TCR is low and dissociates quickly. Tetramerized MHC molecular loaded with candidate peptides promoted the association of TCR with the complex. When labeling with fluorescent dyes, MHC tetramer/multimer could act as staining reagents for epitope-specific T cells, enabling the convenient detection by flow cytometer. Class I and class II tetramer reagents are produced differently and have been used in identifying CD8+ and CD4+ T cells, respectively.

Peptide-MHC Multimer Assay Service at Creative Biolabs

With our deep knowledge of T cell structure and function, Creative Biolabs has developed a series of peptide/MHC molecules. The tetramer and multimer produced using advanced and mature protein design and expression system show high bioactivity and natural structure.

Workflow of Tetramer-Guided Epitope Mapping (TGEM)

TGEM precisely maps antigenic epitopes of known antigens and determines the HLA restrictions.
