Monoclonal Donkey Antibody Discovery Introduction

Monoclonal antibodies are biological macromolecules with a wide range of applications. Compared with polyclonal antibodies, monoclonal antibodies are more suitable for detecting single antigens. Monoclonal Donkey Antibody Discovery

Monoclonal antibodies are antibodies produced by a single clone of B cells and can bind to specific antigens. Monoclonal antibodies are usually more specific and have higher affinity than polyclonal antibodies, and therefore have been widely used in biomedical research and therapy. Donkeys are commonly used as experimental animals, and their immune systems can produce a variety of antibodies. By immunizing donkeys with antigens, polyclonal antibodies can be obtained. However, polyclonal antibodies usually have lower specificity and affinity and are therefore not suitable for some applications. In contrast, monoclonal donkey antibodies have high specificity and affinity and may be a better choice in some cases.

A study showed that anti-distemper virus IgG prepared from donkey serum had significant antiviral effects against canine distemper virus (CDV). In addition, donkey, as a member of large mammals from a wide range of sources, can produce more antibody products to treat canine infectious diseases. This can greatly reduce the cost of treatment and make this treatment more popular. In passive immunotherapy using antibodies from heterologous animals, it is necessary to consider the immune rejection of heterologous proteins by the receptor. The results of this study showed that although there were no strong toxic side effects of treating canine distemper disease with donkey-derived IgG, there were some adverse reactions. The authors hypothesized that these reactions may be related to IgG purity and immune rejection of heterologous IgG by the receptor. To solve these problems, the preparation conditions should be optimized to improve the quality of IgG. In addition, the IgG should be modified. An effective method commonly used today is to treat IgG with papain to cut off the Fc fragment of IgG and prepare the Fab fragment of IgG, thus reducing the antigenicity of IgG. In order to improve the therapeutic effect of donkey-derived anti-CDV IgG, IgG can be chemically modified. Chemical modification cross-links certain cytokines (or chemokines) to stimulate or recruit immune cells to directly kill infected CDV.

Antibody preparation by phage display technology is a commonly used technique for antibody preparation. The phage display library construction can mimic the process of natural antibody library formation without the need for a tedious and time-consuming immunization process. This greatly improves the efficiency of monoclonal antibody preparation and provides a powerful screening capability against arbitrary antigens or even unknown antigens. Generally, commonly used antibodies can be obtained from natural or semi-synthetic antibody libraries. This is the important and most frequently used method to obtain antibodies for most phage display techniques. However, some experiments and studies have higher requirements for antibodies. In many cases where antibody specificity and affinity are required, immune antibody libraries can be constructed. Immune antibody libraries can be enriched by antigen-specific antibodies, and a large percentage of the libraries encode functional antibodies that recognize specific immunogens.
