Neddylation-Specific Antibody Introduction

Neddylation is a protein post translational modification (PTM) similar to ubiquitination. Neddylation has been found to regulate a wide range of cellular activities and is relevant to the pathological processes of liver diseases, metabolic diseases, immune disorders, cancer, and many other diseases. Therefore, protein neddylation has become a popular target for drug discovery. Neddylation Modification

Neddylation is the process by which ubiquitin-like protein 8 (NEDD8) is coupled to its target protein, similar to ubiquitination. The sequences of NEDD8 and ubiquitin are reported to be 59% identical, making neddylation a PTM that resembles the process of ubiquitination. Neddylation occurs through the activation of neural precursor cell-expressed, developmentally down-regulated NEDD8 by three enzymes: an activating enzyme, a binding enzyme, and a ligase. The process is that the NEDD8 precursor is cut off at the C-terminus by a maturation enzyme, exposing Gly76, which makes the NEDD8 precursor active and functional. The NEDD8 activating enzyme catalyzes its attachment to the binding enzyme E2. It is then linked to the lysine of the target protein, catalyzed by NEDD8 ligase E3. The primary role of neddylation is to alter protein function, not degradation.

The most understood substrate for neddylation is the cullin family, which serves as the scaffolding protein for the cullin-RING ligase (CRL) complex (there are three types of ubiquitin ligases E3, one of which is the RING type). CRL is the largest family of RING types and requires multiple proteins to form a complex. Cullin is responsible for linking some of these components together. There are nine human Cullin proteins, most of which require neddylation to be active. Therefore, NEDD8 promotes ubiquitination by activating cullin. CRL controls many important biological processes, including cell cycle progression, DNA repair, signal transduction, gene transcription, embryonic development, genome integrity, and tumorigenesis. In addition, NEDD8 has multiple substrates that control a variety of biological processes. For example, neddylation can inhibit the anti-cancer protein p53, activate the oncoprotein MDM2, and regulate the activity of important molecules such as EGFR, Parkin/PINK1, HIF1α/HIF2α, caspases/IAPs/RIP1, and histone H4. Therefore, neddylation is an extremely valuable drug target. In turn, neddylation modulators are effective drug candidates. Currently, an inhibitor of the NEDD8 activating enzyme NAE, the small molecule compound MLN4924, has entered clinical trials for antitumor.

Especially in the treatment of tumors, NEDD8-activating enzyme inhibitors have demonstrated excellent anti-tumor effects. Several NEDD8 activase inhibitors have been approved and marketed for the treatment of tumors. Since neddylation has many activities and functions, the study of neddylation is essential. In neddylation studies, protein studies are always inevitable. Therefore, neddylation-specific antibodies occupy an important position.
