TACSTD2
Tumor associated calcium signal transducer 2 (TACSTD2) is a 323-amino-acid membrane located glycoprotein containing one thyroglobulin type-1 domain. It belongs to the EPCAM family with a blocked N-terminus. TACSTD2 is also known by other acronyms such as EGP1 (epithelial glycoprotein-1), GP50, M1S1, EGP-1, TROP2, GA7331, or GA733-1. It is encoded by an intronless gene and the protein is regarded as a carcinoma-associated antigen in placenta and pancreatic carcinoma cell lines. TACSTD2 is involved in calcium signal transduction and a mutation in the TACSTD2 is usually associated with gelatinous drop-like corneal dystrophy (GDLD).
Due to its carcinogenic nature and cell surface localization, TACSTD2 has been exploited as a target for ADC-based immunotherapies. It is the target for antibody-drug conjugates such as sacituzumab govitecan, an ADC that bears SN38, a topoisomerase I inhibitor, for accurate cancer elimination.
