Tumor Metabolic Microenvironment
Altering cellular metabolism brings new hope for cancer treatment. The tumor microenvironment is composed of different cell populations and is often characterized by nutrient competition, low pH, limited oxygen, and metabolite accumulation. The demand for bioenergy from rapidly proliferating cancer cells causes cancer cells and immune cells to compete for nutrients needed for anti-tumor defenses. In this case, the tumor itself undergoes metabolic changes, producing an inhibitory immune microenvironment, which in turn inhibits the anti-tumor immune response. And it is worth noting that immune cells that migrate to TME also undergo metabolic changes, develop an immunosuppressive or tolerant phenotype, and promote a metabolism that is more dependent on oxidative phosphorylation and fatty acid oxidation to meet energy requirements. Ultimately, the effectiveness of the anti-tumor immune response of immune cells is disrupted.
