Retinoic Acid-induced Protein 3

Retinoic acid-induced protein 3 (RAI3), also known as RAIG1 or GPRC5, is a protein encoded by the human GPRC5A gene. As a member of type 3-G protein coupling receptor family, it is characterized by seven transmembrane helices, three intracellular loops, three extracellular loops, an extracellular N-terminus, and an intracellular C-terminus. Importantly, RAI3 plays significant roles in multiple biological processes, such as embryonic development, tumorigenesis, inflammation, cellular proliferation, and differentiation. Furthermore, the dysregulation of RAI3 may result in several different types of cancers and other diseases.

Retinoic Acid-induced Protein Marker of Bladder Cancer

RAI3 is an orphan GPRC and has been shown to be differentially expressed in several malignant epithelial cell lines. Down-regulation of RAI3 using siRNA suppressed the growth of some of these cells. Recent studies have demonstrated that it can be used as a potential tumor suppressor or oncogene in various human cancers. For example, RAI3 expression is decreased in non-small-cell lung cancer. However, the level of RAI3 is significantly elevated in gastric cancer tissue which enables it a potential biomarker for this cancer. In addition, the overexpression of RAI3 in hepatocellular carcinoma patients predicts poor prognosis after curative hepatectomy. Most importantly, RAI3 is enriched in urine MPs isolated from individuals with bladder cancer which enables it a promising marker for the diagnosis of bladder cancer.
