Liquid chromatography (LC) is an analytical method that utilizes a liquid mobile phase to separate and analyze components within a mixture. In high-performance liquid chromatography (HPLC), the liquid solvent serving as the mobile phase is pressurized to flow swiftly through the column, facilitating swift and effective separation of the sample components. Mass spectrometer (MS) is an analytical tool used to detect and measure compounds in samples by determining their mass-to-charge ratio (m/z).

LC-MS/MS (liquid chromatography with tandem mass spectrometry) possesses powerful separation and selectivity capabilities. In LC-MS/MS, the sample is initially separated using liquid chromatography. Then, the eluted compounds enter the mass spectrometer, forming charged ions. The tandem mass spectrometer can be divided into MS1 and MS2. In the first mass spectrometer (MS1), ions are isolated and then directed into a collision cell where they undergo fragmentation. The fragmentation process results in the creation of product ions, which are subsequently separated and detected in the second mass spectrometer (MS2).
