CLDN3

The CLDN genes encode a family of proteins involved in the formation and function of tight junction (TJ). As a member of the claudin protein family, CLDN3, also known as claudin3, is the protein encoded by the CLDN3 gene in humans. TJs are critical structures for the regulation of paracellular ion flux and for the maintenance of cellular polarity in endothelial and epithelial. Claudin proteins, which contain approximately 23 members, are important components for the TJs formation. The expression profiles of various claudin proteins are always tissue-specific and even cell type-specific, which means different expression of claudin proteins play specific roles in different tissues or different conditions. It is noteworthy that CLDN3 is normally expressed in the epithelial cells of renal tubules, colon mucosa, mammary gland, bile canaliculi in liver, endothelial cells of the blood-brain barrier, and the acinar epithelial cells of the prostate. In recent years, some studies have shown that CLDN3 is highly increased in ovarian cancer and might be a great biomarker for the diagnosis of ovarian cancer.

CLDN3 Marker of Ovarian Cancer

It has been reported that these claudin proteins are elevated in various cancer types, including breast, uterine, pancreatic, and prostate cancer. The reason why CLSN3 is highly increased in ovarian cancer would be that claudin-3 leads to an increase in motility, invasion, and cell survival of ovarian cancer cells. In addition, claudin-3 is a natural receptor for the Clostridium perfringens enterotoxin, which enables the rapid cytolysis of claudin-3 expressing cells. The recent research shows that CLDN3 can be regulated by the transcription factor Sp1 and through epigenetic modifications including histone modifications and DNA methylation. In summary, claudin-3 may present an exciting new therapeutic target for ovarian cancer and have been served as a great biomarker for the diagnosis of ovarian cancer.
