Mesothelin (MSLN)

Mesothelin (MSLN) was found located on chromosome 16p13.3. It encodes a 71 kDa preprotein which expressed on the surface of mesothelial cells of the pleura, peritoneum, pericardium, and testicular tunica vaginalis. This gene also encodes a preproprotein that is proteolytically proposed to generate two protein products which are megakaryocyte potentiating factor and mesothelin. Mesothelin is a glycosylphosphatidylinositol-anchored protein that may function as a cell adhesion protein. It is also expressed on the cell surface of different tumors including pleural, pancreatic, and especially on the cell surface of ovarian cancers.

Mesothelin Marker of Ovarian Cancer

The studies indicate that mesothelin is a tumor differentiation antigen. These ongoing clinical trials will help define the utility of mesothelin as a target for cancer therapy. A variety of studies have been done to evaluate the function of mesothelin. In Eker rats, the development of tuberous sclerosis-2-induced renal carcinoma was significantly reduced in the absence of a homologue of the MSLN gene. In pancreatic cancer cells, overexpression of MSLN was implicated in significant enhancement of tumor cell growth and migration in vitro. In ovarian cancer models, analyses of correlation between mesothelin expression pathological variability and clinical outcomes indicated that high mesothelin expression was positively associated with chemo-resistance in epithelial ovarian carcinoma patients and short patient survival time. Several studies have indicated important interactions between signaling pathways involved in the development of malignant phenotype and mesothelin. For example, mesothelin was found to induce expression of matrix metalloproteinases 7 (MMP-7) or to enhance expression levels of interleukin 6 (IL-6). Expression of mesothelin is also claimed to confer resistance to apoptosis in response to tumor necrosis factor-alpha (TNF-alpha).
