Error Detection for Protein Modeling

Homology modeling has significant potential as a tool for rational drug design, especially in high-throughput computer selection or simulation strategies. The quality of the final structure depends mainly on the quality of the target and template orientation. However, there are always structural differences between targets and templates, which must be identified and recalculated by ab initio modeling or optimization methods. Therefore, the error detection of modeling is very important for the evaluation of modeling quality. At present, Creative Biolabs’s computational protein design platform has been able to provide error detection services for protein modeling.

Errors in Protein Modeling

The analysis of protein structure provides a basic understanding of most biochemical functions, thus providing a basis for the cause of disease and possible treatment. Since most known protein structures cannot be solved experimentally either technically or sometimes only in time constraints, the molecular model of protein structure is a rapidly developing field. Up to now, the most widely used computational method of protein structure prediction relies on detecting the homology of known proteins and using the protein as a template to simulate the structure of queried proteins. However, as the similarity between the target and the template decreases, the error in the model increases. The errors in the homology model can be divided into five categories:

The evaluation of three-dimensional homology modeling is a difficult task. For method developers and users, assessing the accuracy of methods is critical.

Services

The quality of the prediction model determines the information that can be extracted. Therefore, estimating the accuracy of three-dimensional protein models without known structures is critical to interpreting these models. The model can be evaluated as a whole or as a single area. At Creative Biolabs, our assessment system can automatically evaluate secondary structure prediction, residual contact prediction, folding assignment and comparative modeling of protein structure prediction server. For the error detection of protein modeling, our evaluation mainly focuses on the following aspects:

Our hope at Creative Biolabs is to minimize errors in protein modeling to provide researchers with reliable structural predictions. For more detailed information, please feel free to contact us.
