For initial optimization experiments, transfect suspension cells using 2:1~5:1 ratios of Magic™ Transfection Reagent-Suspension Cells (μL) to DNA (μg), respectively.
For most suspension cell types, the Magic™ Transfection Reagent-Suspension Cells : DNA (2:1) ratio provides excellent transfection level. For some hard-to-transfect cells, the Magic™ Transfection Reagent-Suspension Cells : DNA (5:1) ratio is recommended.
Good cell culture is a prerequisite for successful transfection. Low passage times (<50 division cycles) avoid changes in cell genotypes. We recommend subculture of cells 24 hours prior to transfection to provide normal cell metabolism and increase the likelihood of DNA uptake.
Avoid using serum during complex formation between transfection reagent and plasmid DNA as serum may inhibit complex formation.
Plasmid DNA concentration can be accurately determined by using absorbance at 260 nm; DNA content can be determined by absorbance at 260 nm.
