The vector of anti-B7-H4 chimeric antigen receptor (CAR) is constructed for the engineering of T cells to target Human B7-H4. The T cells are genetically modified through transduction with a lentiviral vector expressing scFv of anti-B7-H4 antibody linked to 4-1BB and CD3ζ signaling domains. And the vector product was designed for the treatment of Ovarian cancer.
CAR Construction : Fig.1 Flow cytometric analysis of either B7-H4 WT/293 or B7-H4 H250Q MT/293 transfectants showed that the stable transfected cell lines were successfully constructed. Shown are mean ± SD values from three independent triplicates. Zhang, L., Wu, H., Lu, D., Li, G., Sun, C., Song, H., ... & Zhang, X. (2013). The costimulatory molecule B7-H4 promote tumor progression and cell proliferation through translocating into nucleus. Oncogene, 32(46), 5347-5358. |
