TCR Tetramer

Peptide-MHC tetramers have been extensively applied for the detection and quantification of antigen-specific T cell populations and research MHC functions. Based on a powerful TCR development platform, scientists at Creative Biolabs have experience in the generation and optimization of TCR. We extend tetramerization technology to TCRs and have the ability to manufacture custom TCR tetramers that bind MHC-peptide complexes with high affinity.

TCR tetramers serve as surface staining reagents for analysis of the MHC-peptide interface and the design of immunotherapeutics and vaccines. Compared to TCR monomers, TCR tetramers have higher affinities and can extend the half-life of TCR:pMHC interactions. TCR tetramers can provide new insight into MHC–mediated antigen presentation.

Preparation of TCR Tetramers

At Creative Biolabs, our team of scientists is dedicated to providing customized TCR tetramers to meet the needs of customers. Provide us with your candidate's TCR sequences or your specific TCR of interest, we can custom manufacture a TCR tetramer based on your needs.

A TCR Tetramer is prepared using biotinylated specific TCRs and fluorophore conjugated streptavidin which has four biotin binding sites.

  • Generation of Soluble TCR Chains
    Soluble TCRs have been expressed in various recombinant protein expression systems. For example, insect cells and bacterial expression systems (E. coli) are widely used to support the expression of the gene of interest. Two polypeptide chains (α and β chains ) of TCR are expressed separately from two distinct vectors and paired by a non-native disulfide bond. A biotinylation target motif is also fused to the C terminus of the TCR β chain to allow tetramerization with extra-virgin.
  • Refolding Soluble Monomeric TCRs
    Purified TCR chains are refolded for several days to obtain soluble TCR monomers.
  • Biotinylation of Monomeric TCRs
    The refolded monomer is labeled with biotin.
  • Multimerization of Refolded TCRs
    TCR monomers are tetramerized by the addition of fluorochrome-conjugated streptavidin to the monomers.
