Humanized mouse models show great promise as pre-clinical research models for CAR-T evaluations. At present, several studies have been undertaken to address the present restrictions associated with the inclusion of humanized mice, which aim to enhance the myeloid in humanized mouse models for more comprehensive human immune system restoration. Myeloid-enhanced Humanized Mice Models

A myeloid-enhanced humanized mouse model that is genetically modified to express human interleukin-3, GM-CSF, and SF on the NSG mouse background. The number of human myeloid cells and human CD4+ Treg cells in the NSG-SGM3 mouse increases. The mouse model has great application potential in detecting cytokine storms caused by CAR-T and supporting the reconstruction of CAR-Mac cells in vivo.
A myeloid-enhanced humanized mouse model was obtained by introducing human cytokines IL-3 and GM-CSF based on NOG. huNOG-EXL is currently the most complete animal model for the reconstruction of the human immune system. It can be used to verify the anti-tumor effect of PD1 antibodies and also supports the validation of drug efficacy in the acute myeloid leukocyte (AML) PDX model.
A myeloid-enhanced humanized mouse model equipped with a BAC transgenic that encodes human SIRPα. It reduces the phagocytosis of human CD47-expressing cells, which allows for extremely lenient human hematopoiesis.
