The vector of anti-CSPG4 chimeric antigen receptor (CAR) is constructed for the engineering of T cells to target Human CSPG4. The T cells are genetically modified through transduction with a lentiviral vector expressing scFv of anti-CSPG4 antibody linked to CD28 and CD3ζ signaling domains. And the vector product was designed for the treatment of Breast cancer.
CAR Construction : Fig.1 WM9, WM35 melanoma cells were subjected to flow cytometry to determine the percentage of CSPG4‑positive cells using the anti‑CSPG4 9.2.27 mAb. Data are representative of triplicates. Uranowska, K., Kalic, T., Valtsanidis, V., Kitzwögerer, M., Breiteneder, H., & Hafner, C. (2021). CAR Construction : Fig.2 CSPG4 wildtype and mutants display similar expression levels in the stable transfected WM1552C cell lines. Flow cytometry was performed using CSPG4 antibody 9.2.27 and normal mouse IgG2a as a negative control. Yang, J., Price, M. A., Wanshura, L. E. C., He, J., Yi, M., Welch, D. R., ... & McCarthy, J. B.
