The vector of anti-EGFR chimeric antigen receptor (CAR) is constructed for the engineering of T cells to target Human EGFR. The T cells are genetically modified through transduction with a lentiviral vector expressing scFv of anti-EGFR antibody linked to CD28 and CD3ζ signaling domains. And the vector product was designed for the treatment of Melanoma.

CAR Construction : Fig.2 MM-151 is superior to cetuximab at blocking tumor growth of a high-affinity EGFR ligand–expressing murine xenograft model. Xenograft tumors of H322M-HB-EGF cells were established in immunecompromised mice and treated with PBS, indicated doses of cetuximab, or indicated cetuximab-equivalent doses of 25E-Trio, and tumor volumes were measured by calipers.
