Importance of chemoselective ligation reactions
Chemoselective ligation reactions are highly effective and specific covalent bond forming reactions which are capable of proceeding within a physiological environment. Commonly, chemoselective ligation reactions could create long peptides or protein chains and generate a unique product. In these reactions, two mutually reactive functional groups undergo a chemoselective reaction independent of all the unprotected functional groups borne by the amino acid side chains. It is similar to enzymatic reactions with exquisite specificity. In addition to the high specificity, chemoselective ligation reactions show important advantages, which allow a broader range of partners to perform reactions. Because of the unique characteristics, chemoselective ligation reactions have been used in many applications, such as carbohydrate-based structure construction, cell-surface engineering, generation of glycoproteins/peptides and glycosylated natural products.
